The fighting broke out when members of the Revolutionary Armed Forces of Colombia (FARC) detonated an explosive device in a village in the Colombian southwestern province of Cauca on Monday.
The clashes, which continued on Tuesday, have also left 11 people injured and 750 displaced.
Colombian Interior Minister German Vargas has described the situation in the area as “complicated.”
Earlier this month, the rebel group orchestrated a similar attack in a nearby town, killing seven people and injuring many others.
FARC, Latin America’s oldest rebel group, has been battling the Colombian government since 1964.
The rebel organization is believed to have an estimated 8,000 fighters operating across a large swathe of the eastern jungles of Colombia.
In recent years, however, the US-backed Colombian military has pushed the group out of their encampments around major cities and into the dense jungles and high peaks of the Andean mountains.
The Colombian security forces have also killed a number of FARC commanders and arrested many others.
The group is struggling to gain control over regions with Pacific coastlines — key for exporting narcotics and smuggling weapons.
